Nuclear magnetic relaxation spectroscopy in solutions of bovine hemoglobin.

چکیده

The longitudinal proton relaxation time Tt of aqueous solutions of bovine ferrihemoglobin and carboxilated hemoglobin was measured in the Larmor frequency range 3-103 Hz to 1,2 -108 Hz. The dispersion curves are explained by diamagnetic and electron paramagnetic relaxation mechanisms. An efficient direct dipolar coupling between the paramagnetic heme-groups and the water protons in ferrihemoglobin solutions can be excluded. The flat dispersion behaviour of the diamagnetic contribution suggests a broad distribution of molecular motions i. e. a certain flexibility of the hemoglobin molecule.

Quantitative determination of methemoglobin by measuring the solvent-water proton-nuclear magnetic resonance relaxation rate.

We report a new method for the quantitative determination of human methemoglobin (metHb) based on the measurement of the solvent-water proton-nuclear magnetic resonance (NMR) relaxation rate R1 [normalized to 1 mmol/L hemoglobin (Hb) concentration]. MetHb (%) is estimated from the linear dependence of R1 on the metHb concentration, taking into account the simple relationship [MetHb] = [(R1 - R1...

Applications of natural-abundance nitrogen-15 nuclear magnetic resonance to large biochemically important molecules.

Natural-abundance nitrogen-15 nuclear magnetic resonance spectroscopy of enzymes and other biopolymers is found to be feasible using newly available instrumentation. The long correlation times of such molecules result in short spin-lattice relaxation times, and these in turn allow rapid signal accumulation. The advantages of short T1 values are sometimes offset, however, by unfavorable nuclear ...

Characterizing monoclonal antibody formulations in arginine glutamate solutions using 1H NMR spectroscopy

Assessing how excipients affect the self-association of monoclonal antibodies (mAbs) requires informative and direct in situ measurements for highly concentrated solutions, without sample dilution or perturbation. This study explores the application of solution nuclear magnetic resonance (NMR) spectroscopy for characterization of typical mAb behavior in formulations containing arginine glutamat...
